abstract | The invention provides a combined reagent A for determining the total bile acid in serum and a detecting method. The combined reagent A comprises a first reagent and a second reagent, wherein the first reagent consists of Tris-HCl (hydrogen chloride) buffer liquid, 3 alpha-HSD (3 alpha-hydroxysteroid dehydrogenase) and coenzyme (NAD(nicotinamide adenine dinucleotide)<+>or NADH (nicotinamide adenine dinucleotide)), and the second reagent consists of diaphorase, 2,6- DCPIP (dichloropheno lindophenol) and Tris-HCl buffer liquid. The blue DCPIP is constantly reduced into colorless products through coenzyme circulation, and the bile acid concentration is quantificationally detected. The coaxial cospinning electrostatic spinning technology is also utilized, the 3 alpha-HSD, thediaphorase and the coenzyme are subjected to in-situ embedding into a hollow polymer nanometer fiber cavity, and a combined reagent B in an immobilized form is prepared. The DCPIP is added into bile acid containing serum samples to be tested, then, the immobilized combined reagent B is added, the reaction starts, and the bile acid detection is carried out. The immobilized combined reagent has the obvious advantage of high storage stability, and application prospects are realized in the aspect of clinically, accurately and fast determining the bile acid. |
