titleOfInvention In vitro screening method of serum nucleic acid aptamers by multi-step concatenating selection
abstract The invention discloses an in vitro screening method of serum nucleic acid aptamers by multi-step concatenating selection, belonging to the technical field of biomedical detection and analysis, characterized by eliminating individual difference by using a mixed serum as a screening target, conducting reverse screening by using a normal serum to remove nucleic acid sequences capable of combining with non-specific serum components from a library, then conducting screening by using the mixed serum to clone, sequence and separate nucleic acid aptamers, and selecting the nucleic acid aptamers having large application value by carrying out structural simulation analysis, singularity analysis, affinity analysis, and application value analysis on the nucleic acid aptamers. According to the invention, interference of large amounts of non-specific components in the serum is overcome, the method can directly use the serum as the target to screen out the nucleic acid aptamers having disease-specific components from the serum with high through-put, has the advantages of efficiency and economy, can lay the foundation for establishing analysis technology based on nucleic acid aptamers, systematically exploring the rules of the occurrence and development of diseases, and acquiring bioinformation related to clinic diagnosis and treatment, and has good application value.
