abstract | The invention discloses a system for amplifying an alkaline phosphatase label signal. Alkane, olefin or alkyne alcohol phosphate is used as a substrate and generates free phosphoric acid and corresponding alcoholic compounds under the action of alkaline phosphatase; the alcoholic compounds generate aldehyde or ketone compounds and hydrogen peroxide (H2O2) under the participation of oxygen and under the action of alcohol oxidase; the generated aldehyde or ketone compounds are reduced into alcoholic compounds under the action of alcohol dehydrogenase in the presence of NADH (Nicotinamide Adenine Diuncleotide Hydrogen); and the generated alcoholic compounds continuously participate in a circulating reaction. A large amount of H2O2 which is circularly generated is matched with different substrates to be catalyzed by peroxidase and then the catalyzed substance can be detected by a colorimetric method, an electrochemical process, a chemiluminescence method or a fluorimetric method. Therefore, according to the system, independent alkaline phosphatase or alkaline phosphatase connected with an antigen, a hapten, an antibody, protein, nucleic acid and oligonucleotide can be sensitively detected and the function of amplifying the signal is achieved.
