abstract | The invention relates to the biological technical field, which discloses an amplification primer for detecting a drug-resistance mutation site of an ABL kinase domain of a BCR/ABL fusion gene. The upstream primer of the amplification primer has a nucleotide sequence shown in SEQ ID No. 1 or SEQ ID No. 2, and the downstream primer of the amplification primer has a nucleotide sequence shown in SEQ ID No. 3. The invention also discloses a method and kit for detecting a drug-resistance mutation site of an ABL kinase domain of a BCR/ABL fusion gene. The invention amplifies the BCR/ABL fusion gene of patients with chronic myelogenous leukemia (CML) by one-step polymerase chain reaction (PCR) amplification, and carries out sequencing by using a specific sequencing primer; and can detect more than twenty kinds of common drug-resistance mutations of the ABL kinase domain of the BCR/ABL fusion gene in a reaction system, which lowers experiment cost, greatly improves accuracy and sensitivity of gene sequencing results, and has a large-scale application prospect. |
