abstract | The invention discloses a recombinant lysostaphin locus specific integration vector and a recombinant cell established by same. Recombinant lysostaphin gene consists of cow growth hormone signal peptide and mutant lysostaphin mature peptide, regulatory sequences of casein gene are added to the 5' end and the 3' end of the recombinant lysostaphin gene, and an insulator element of chicken beta lactoglobulin is added behind the casein 5' end and a terminator codon of enhanced green fluorescent protein (EGFP) respectively. A bovine fetal fibroblasts cell integrated with target gene is used as a donor cell for constructing transgenic cloned embryo through nuclear transfer, and a transgenic cloned embryo is obtained through somatic cell nuclear transfer (SCNT). By the normally developed transgenic cloned embryo, mastitis-resistant high-yield cow with the transferred recombinant lysostaphin gene is expected to be produced through an embryo transplantation technology. |
