abstract | The invention relates to a synthetic method of erlotinib hydrochloride. It has the following process flow: a. Use m-nitrobenzaldehyde as raw material to synthesize m-nitrocinnamic acid; b. Add bromine to obtain 2,3-dibromo-3-(3'-nitrophenyl)propionic acid; c. .Decarboxylation and dehydrobromination to obtain (Z)-β-bromo-(3'-nitrophenyl)-ethylene; d. Reaction with metal hydride to obtain m-nitrophenylacetylene; e. Reduction to obtain m-aminophenylacetylene ; f. Reaction with 4-chloro-6,7-bis-(2-methoxyethoxy)-quinazoline to obtain erlotinib hydrochloride. The method has cheap and easy-to-obtain raw materials, mild reaction conditions, simple operation, high yield, and is suitable for scale-up. |
