abstract The invention provides a tag used for multiple nucleic acid sequencing and an application method thereof. Based on an existing Illumina/Solexa sequencing platform provided by the Illumina Company, aiming at a sequencing library, especially an Illumina/Solexa sequencing sample library creating method, tags with different lengths, especially unique gradient tag sequences (index 1-6), are designed. The tags are embedded into the sequencing library through connectors, especially in a 3' connector of the Illumina/Solexa sequencing library, a sequencing tag library is successfully created. Especially the library creating method of the Illumina/Solexa sequencing tag library is suitable for creating any biological sequencing tag library, and is successfully used in Illumina/Solexa sequencing, thus not only increasing the sequencing flux of Illumina/Solexa sequencing samples, but also reducing the cost of Illumina/Solexa sequencing.
