abstract | The invention provides a novel preparation method of a recombinant antibacterial polypeptide medicament, and relates to the production technology of recombinant antibiotic medicament. The method comprises the following steps: (1) preparing an escherichia coli strain containing recombinant plasmids, and carrying out cryopreservation; (2) propagating the strain by adopting a liquid production culture medium; (3) performing inducible expression to the recombinant antibacterial polypeptide, and separating and purifying the recombinant antibacterial polypeptide. The method is characterized in that: the final solution of the liquid production culture medium comprises the following components in percentage by mass/volume: 0.4 to 0.7 percent of disodium hydrogen phosphate, 0.1 to 0.6 percent of monopotassium phosphate, 0.05 to 0.2 percent of ammonium chloride, 0.0005 to 0.001 percent of calcium chloride, 0.5 to 2.5 percent of magnesium sulfate, 1 to 3 percent of peptone, 0.5 to 1 percent of yeast powder, 0.1 to 0.5 percent of glucose, 0.2 to 0.8 percent of calcium chloride, and the balance of water. The invention further optimizes the method in the aspects of propagating parameters, and is applicable to large-scale antibacterial peptide production; and the prepared antibacterial polypeptide has high purity. |
