abstract | The invention relates to a colibacillus containing alpha-amino-acid ester hydrolase gene, which solves the problem of hydrolytic action of beta-lactamase of the original host bacterium Xanthomonas rubrilineans on beta-lactam ring. The invention determines the complete sequence of alpha-amino-acid ester hydrolase gene derived from the Xanthomonas rubrilineans strain and a zymoprotein coded by the same, conforms the target gene aeh, and provides a recombinant plasmid pET28a containing the gene and constructed E.coliBL21(DE3) host bacterium. The Latin name of the host bacterium is Escherichiacoli, the preservation unit is the Common Microorganism Center of China Committee of Culture Collection for Microorganisms, the preservation date is April 11th, 2011, and the preservation number is CGMCCNO.4757. The amino acid sequence of the target gene is disclosed as Sequence 2 in the sequence table, and the nucleic acid base sequence of the target gene is disclosed as Sequence 1 in the sequence table. The host bacterium overcomes the defect that the original host bacterium beta-lactamase has a damaging effect on many cephalosporin antibiotic mother nuclides, such as 7-ADCA and the like and derivatives thereof. The host bacterium can be generally used for synthesizing cefalexin, cefprozil, cefaclor, cefatrizine and cephazoline. |
