The method includes the following steps: spirulina cells are frozen under negative 20 DEG C for 1.5 hours and then thawed under 4 DEG C for 1 hour, and the freezing and melting process is repeated for five times until the spirulina cells are completely broken; Tris-HCL extracting solution is used for carrying out extraction for three times to obtain water-soluble protein, and SDS (sodium dodecyl sulfate) extracting solution is used for extracting water-insoluble protein from precipitate. Compared with the conventional spirulina 2-DE (dielectrophoresis) protein preparation method, the spirulina 2-DE protein preparation method established by the invention, i.e. a step-by-step freeze-thaw wall-breaking purification method, is easy to operate, has the remarkable advantages of convenient and simple operation, simple instruments and equipment, low cost, richer information quantity and the like, and can remarkably increase the information quantity and resolution of a spirulina protein 2-DE map, thus laying a necessary foundation for the subsequent bioinformatic researches, such as a proteomics research.
