abstract The invention relates to a 'spore-free crystal-free bacterial strain of Bacillus thuringiensis as well as a construction method and applications thereof', belonging to the technical field of biology. According to the invention, through comparing the transcriptional activities of four cry gene promoters (PcrylA, Pcry3A, Pcry4A and Pcry8E) with different transcription mechanisms of Bacillus thuringiensis in a spore-free mutant HD-delta SpolllD, a promoter of a high-activity promoter cry8E gene of the appropriately expressed foreign gene of the bacterial strain is screened, and a foundation for establishing a foundation for constructing an efficient expression system is established, and meanwhile, an expression element for the development of novel pesticides is provided. On the basis, a spore-free crystal-free mutant HD-delta SpolllD is constructed through a high-temperature inducing method; in the bacterial strain, the CrylAc protein expression quantity instructed by the four cry gene promoters with different transcription mechanisms is compared, and is consistent with the protein expression condition in the HD-delta SpolllD, which shows that the spore-free bacterial strain HD-deltaSpolllD and the spore-free mutant HD-delta SpolllD both can be used as the genetic materials for analyzing the foreign cry gene expression.
