abstract The invention provides a method for testing the contents of absorbed and transported diterpene alkaloids of radix aconiti carmichaeli in the Caco-2 cell model. The method includes the following steps: (a) the diterpene alkaloids in radix aconiti carmichaeli are extracted, so that alkaloid extract is obtained; (b) the alkaloid extract is dissolved in Hanks balanced salt buffer solution, absorption and transportation are carried out in the Caco-2 cell model by using the obtained solution as a supply side and the Hanks balanced salt buffer solution as a receiving side, and a sample to be tested is obtained from the receiving side; (c) with reserpine as an internal standard, an ultrahigh-performance liquid chromatograph and a mass spectrograph are adopted to separate and test the sample to be tested, so that a mass spectrogram is obtained; (d) according to the mass spectrogram and a preestablished standard working curve, the content of each absorbed and transported alkaloid in the Caco-2 cell model is obtained. The ultrahigh-performance liquid chromatograph has a good separation effect on complex samples, while the mass spectrograph has higher sensitivity and accuracy, therefore the testing result is more accurate.
