abstract | The invention discloses a gene for improving the tolerance of microbial organic solvent as well as a coding protein and application of the gene and belongs to the technical field of biological engineering. The mmsB gene from P.putidaCCTCCNO:M2011442, which is disclosed by the invention, has sequence length of 888bp and 295 coded amino acids; and hydroxy-3-methylpropionic acid-dehydrogenase coded by the gene has the function of promoting the tolerance of the microbial cell organic solvent. Proved by two-dimensional electrophouesis and thallus solvent resistance experiments, the improvement on the expression level of the mmsB gene in the P.putida is related with the improvement on the tolerance of the organic solvent of the mmsB gene; meanwhile, the tolerance of organic solvent of Gram-negative bacteria, such as Escherichia coli cells can be remarkably improved by recombinant expression of the mmsB gene in Escherichia coli host. The invention provides an effective method for explaining the tolerance of the microbial cell organic solvent and provides theoretical basis for constructing a solvent-tolerant integral cell biocatalyst for industrialization. |
