abstract A rooting transplanting method for tissue culture poplar seedlings is used for solving the problem that the existing rooting transplanting methods lead to low survival rate of the tissue culture poplar seedlings. The rooting transplanting method for tissue culture poplar seedlings includes: firstly, placing tissue culture poplar seedlings into test tubes containing liquid rooting medium, erecting the tissue culture poplar seedlings, covering openings of the test tubes with seal films, allowing the seedlings to root to obtain the rooted seedlings; secondly, moving the rooted seedlings with 12-14mm long roots to a container with distilled water from the test tubes, and covering the rooted seedlings with a transparent container to preserve moisture for water culture; and thirdly, transplanting the rooted seedlings subjected to water culture to soil, covering the rooted seedlings with a transparent container to preserve moisture for 16 days, removing the transparent container, transplanting the rooted seedlings with the soil into a greenhouse and completing rooting and transplanting of the tissue culture poplar seedlings. The rooting transplanting method is safe and reliable, and the transplant survival rate of the tissue culture poplar seedlings transplanted into the greenhouse can reach more than 95%.
