abstract The invention discloses a method for determining the content of pectin. The method comprises the following steps: 1) acquiring a sample to be determined; 2) adding a trifluoroacetic acid solution of which the concentration is 0.5-5mo1/L in the sample to be determined in the step 1), so as to obtain a hydrolysate of the sample to be determined; 3) carrying out drying processing on the hydrolysate of the sample to be determined by a N2 drying manner, adjusting pH value to neutral and fixing the volume by water; 4) weighing galacturonorhamnan as a standard substance; and preparing into a gradient standard solution of which the concentration is 0.001mo1/L-0.01mo1/L; 5) respectively carrying out 1-phenyl-3-methyl-5-pyrazolone derivation on the matter obtained in step 3) and the gradient standard solution obtained in the step 4); 6) determining a derivation sample by adopting a high performance liquid chromatography; and 7) making a standard curve taking the standard galacturonorhamnan concentration as a horizontal coordinate and peak area as a vertical coordinate; and inspecting the corresponding concentration of the sample to be determined on the standard curve according to the peak area, so as to calculate the content of the pectin in the sample to be determined.
