abstract | The present invention relates to a use of an AMP-activated protein kinase (AMPK) activator for relieving and treating metabolic diseases wherein AMPK is an energy sensor and key enzyme for regulating lipid and glucose metabolism in a living body. When AMPK is activated, synthesis of lipid and cholesterol is inhibited; body fat is decreased; lowering of blood glucose is facilitated; and obesity, diabetes, and hyperlipidemia are relieved. In the present invention, novel compounds Damulin A (2alph,3belta,12belta-trihydroxydammar-20(2)-E,24-diene-3-O-[belta-D-glucopyranosyl-(1)-belta-D-glucopyranoside]) and Damulin B (2alpha,3belta,12belta-trihydroxydammara-20,24-diene-3-O-[belta-D-glucopyranosyl-(1)-belta-D-glucopyranoside]) are used as the AMPK activator for the active ingredients for relieving and treating metabolic diseases including obesity, diabetes, and hyperlipidemia. In addition, the present invention provides a method for increasing the amount of Damulin A or Damulin B which is the valid indicator for AMPK activation by treating Gynostemma pentaphyllum extracts at high temperature and high pressure. Therefore, the present invention also provides a use of novel Gynostemma pentaphyllum extracts with remarkably increased capacity of activating AMPK for relieving or treating metabolic diseases such as obesity, diabetes, and hyperlipidemia. |
