abstract | The invention belongs to the technical field of medicinal preparations, and discloses an osmotic-pump timing sustained and controlled release capsule and a preparation method thereof. The capsule comprises a heterogeneous membrane capsule body, a non-water permeable capsule cap, and contents loaded inside. The contents comprise a pharmacologically active substance, a suspension and a penetration enhancer, and the weight percents are: 5%-30% of pharmacologically active substances, 40%-80% of suspensions, and 10%-50% of penetration enhancers; the heterogeneous membrane capsule body is an asymmetric bilayer membrane shell prepared by capsule material liquid and quenching liquid; the non-water permeable capsule cap is prepared by filling capsule liquid into a mold and volatilizing the liquid; the contents are directly loaded inside in a form of powder or particles; the capsule cap and the capsule body are sealed by the capsule material liquid; and a hole with a diameter of 0.3-0.5 mm is drilled on the top of the capsule cap. The capsule of the invention can be used to treat diseases with the characteristic of chronopharmacokinetics, and the system releases drugs with a constant speed by using osmotic pressure as power, which effectively avoid the peak-valley phenomenon of plasma concentration in the body, reduce side effect of the drugs, and improve patient compliance. |
