abstract | The invention discloses a method for purifying inclusion body protein, comprising the following steps: inducing inclusion body protein through IPTG and carrying out centrifugation at the temperature of 4 DEG C, directly purifying the medium supernatant; carrying out ultrasonic cracking and centrifugation on a cell mass, abandoning the supernatant, adding an 1*bind buffer and suspending the inclusion body mass, carrying out ultrasonic cracking and centrifugation, adding the supernatant to an Ni chromatography column, washing the effluent with 3 volumes of the 1*bind buffer washing column bed and 3 volumes of a Wash buffer washing column bed; successively adding four renaturation buffers containing arginine in the Ni chromatography column; then adding an Elute buffer to the Ni chromatography column and eluting the target protein; taking and adding the purified medium supernatant in a dialysis bag, carrying out dialysis for 2 h at the temperature of 4 DEG C with PBS, and carrying out centrifugation at the temperature of 4 DEG C for 15 min and settling the target protein. The method is rapid and simple, and reduces production cost. According to the invention, the target protein with biological activity can be effectively acquired. |
