abstract | The invention discloses a liquid phase chip for the polymorphic detection of a fat mass and obesity associated (FTO) gene. The polymorphic detection liquid phase chip comprises a wild type specific allele specific primer extension (ASPE) primer and a mutant type specific ASPE primer which are designed aiming at polymorphic loci of the FTO gene, microspheres which are wrapped with specific anti tag sequences and a primer for amplifying a target sequence containing mutant loci, wherein each ASPE primer consists of a tag sequence at the 5' end and a specific primer sequence at the 3' end; the specific primer sequences are selected from SEQ ID No. 27 to SEQ ID No. 52; the tag sequences are selected from SEQ ID No.1 to SEQ ID No. 26. The invention also discloses specific primers for the polymorphic detection of the FTO gene. The coincidence rate of a detection method provided by the invention and a sequencing method is up to 100 percent. The prepared polymorphic detection liquid phase chip for the FTO gene has a high signal noise ratio, and cross reaction is not formed between a designed probe and the anti tag sequences basically. |
