titleOfInvention Method for preparing sterile rose vine for tissue culture
abstract The invention relates to a rose tissue culture technique and aims to provide a method for preparing a sterile rose vine for tissue culture. The method comprises: trimming and washing a rose vine, soaking the rose vine in phenylthiocarbamide (PTC) aqueous solution at a concentration of 25 percent (v/v), and washing with distilled water; filling the rose vine forward and vertically into a tissue culture bottle which is sterilized by high-pressure steam, covering the bottle cap, placing the bottle in a 37 DEG C constant-temperature tank for 24 hours, transferring the bottle to a 4 DEG C refrigerator for 48 hours and repeating the step once; and cutting the rose vine into small sections with axillary buds under a sterile condition, sterilizing by soaking and in 70-percent alcohol and by shaking, taking the small sections out, washing the small sections with sterile water, soaking in 0.002 liter of mercury for 9 minutes, taking the small sections out, washing with sterile water, and obtaining the sterile rose vine for tissue culture. With the PTC aqueous solution and alternating treatment at 37 DEG C and 4 DEG C, the method has the advantages of higher sterile effect, higher germination rate, simpler and more convenient operation, time and labor conservation, lower cost, higher production application performance and the like compared with the conventional method.
