abstract | The invention discloses a liquid chip for detecting KRAS gene mutation. The liquid chip mainly comprises: (A) respectively designed wild type and mutant ASPE primer pairs aimed at KRAS gene mutation sites; (B) microspheres respectively coated with specific anti-tag sequences, wherein the microspheres have different color codes, and the anti-tag sequences can be complementarily coupled with selected tag sequences; (C) amplimers used for producing KRAS gene target sequences containing corresponding mutation sites of Codon 12, Codon 13 and/or Codon 61 through amplification. The result obtained with the detection method provided by the present invention has a coincidence rate reaching 100% with that of a sequencing method. The prepared liquid chip for detecting KRAS gene mutation has excellent signal to noise ratio. Basically no cross reaction occurs between designed probes and the anti-tag sequences. With the liquid chip provided by the present invention, parallel detections of various mutants on a same site and various mutation sites can be realized. |
