titleOfInvention Preparation method of body protection polypeptide (BPC 157 peptide)
abstract The invention discloses a method for synthesizing body protection polypeptide (BPC 157 peptide), comprising the following steps of: with any one of trityl chloride resin, 4-methyl-trityl chloride resin, 4-methoxyl-trityl chloride resin, 2-chloro-trityl chloride resin and Wang resin as a starting raw material, carrying out peptide coupling reaction by using one of DIC/HOBt (Hydroxybenzotrizole), DIC (N,N-diisopropylcarbodiimide )/HOAt (1-Hydroxy-7-azabenzotriazole), BOP (Benzotriazole-1-yl-oxy-tris-(dimethylamino)-phosphonium hexafluorophosphate)/HOBt, BOP/HOAt, HBTU (O-Benzotriazole-N,N,N',N'-tetramethyl-uronium-hexafluoro-phosphate )/HOBt, HBTU/HOAt, HATU ((2-(7-Aza-1H- benzotriazole-1-yl)-1,1,3,3-tetramethyluronium hexafluorophosphate) /HOBt, HATU/HOAt and HCTU (5-Chloro-1-[bis(dimethylamino)methylene]-1H-benzotriazolium 3-oxide hexafluorophosphate)/HOBt as a condensing agent to obtain protective body protection polypeptide (BPC 157 peptide) resin; then simultaneously carrying out side chain protective group removal and peptide cutting to obtain a BPC 157 peptide crude product; and then separating and purifying with a C18 (or C8) column, freezing and drying to obtain the body protection polypeptide (BPC 157 peptide) pure product. In the method, microwave radiation can be adopted to speed up reaction, shorten synthesis period of polypeptide and reduce production cost. The body protection polypeptide (BPC 157 peptide) prepared by adopting the method disclosed by the invention has the advantages of available raw materials and auxiliary materials, stable process, high peptide coupling yield, controllable quality, high yield, high purity, low production cost, short period, mass production and the like and completely meets application requirement of the body protection polypeptide (BPC 157 peptide).
