abstract The invention discloses a method for the high density culture of haemophilus parasuis for preparing vaccines, comprising the following steps: A, flat-activating freeze-dried seed strain 4, 5 and (or) 13 by using TSA for 10-16h until single colonies grow; B, selecting 3-6 single colonies to put in a TSB shake-flask first seed culture medium, cultivating for 8-14h until OD600 value reaches 0.4-0.6; C, transferring the first seed liquid into a second shake-flask culture medium with the inoculation amount of 1-5% V/V, cultivating for 8-12h up to exponential phase; and D, transferring the second seed liquid into a fermentation-cylinder fermentation medium with the inoculation amount of 1-3% V/V, controlling PH in 6.8-7.2 and OD in 40-60% to maintain the microbe logarithmic growth, when PH value becomes stable and dissolved oxygen rises, taking a pot to collect the bacteria to obtain the somatic antigen of haemophilus parasuis with high density. The method has the advantages of wide raw material source, lost cost, fast microbe growth and high density, and is easy to realize the large-scale high density culture of somatic antigen of haemophilus parasuis.
