abstract | The present invention relates to a new method for synthesizing L-desmethylphencyclononate. The present invention specifically relates to the method for preparing the left-handed demethylphencyclononate represented by formula I or a salt thereof, Formula I which comprises the following steps: (i) in the presence of a suitable reagent in a suitable solvent, make (R)-α-phenyl-α-cyclopentyl-α-glycolic acid methyl ester and N-benzyl-3- Azabicyclo-[3.3.1]-nonan-9-ol was transesterified to give (R)-α-cyclopentyl-α-phenyl-glycolic acid-9-[N-(benzyl)-3- Azabicyclo (3.3.1) nonyl] ester; (ii) in the presence of a suitable catalyst in a suitable solvent, make (R)-α-cyclopentyl-α-phenyl-glycolic acid-9-[N-( benzyl)-3-azabicyclo(3.3.1)nonyl] ester is subjected to catalytic hydrogenation to remove the benzyl group to give a compound of formula I; and optionally (iii) salt-forming the compound of formula I in the presence of an acid to give Salts of compounds of formula I. The present invention also relates to the preparation method of the key intermediate used in the above method. The inventive method has the advantageous features described in the present invention. |
