abstract The invention discloses a method for obtaining genetically modified sea island cotton. The method comprises the following steps: preparing corresponding agrobacterium fluid, centrifuging and recovering the agrobacterium fluid, and adding a 1/2 MS (Murashige and Skoog) culture medium, 20% of cane sugar, 0.01% (v/v) of Selwet and 0.01% (v/v) of DMSO (dimethylsulfoxide) to ensure the OD (optical identity) value to reach around 0.6; placing the mixture in a small sprinkling can and spraying a sea island cotton flower which unfolds in the morning; after spraying, covering the flower with a paper tube and marking the flower; after seeds become ripe, planting a transformation T0 substitute to carry out gene marker detection and PCR (polymerase chian reaction) detection. The method has the advantages of simplicity in operation, freedom from restriction of genotype, high transformation efficiency, reduced workload and capabilities of promoting researches on gene transformation of the sea island cotton and accelerating molecular breeding process. At present, no reports on transformation of the sea island cotton agrobacterium by the infection method could be found.
