abstract | The invention relates to a gene methylation state detection kit used for predicting the course of severe hepatitis. The kit contains a reagent used for performing hydrosulfite modification to the genomic DNA, a primer pair aiming at the methylation specificity and non-methylation specificity of the human glutathione S-transferase M3 promoter and 2*Taq PCRMasterMix. The detection method comprises the following steps: after hydrosulfite modification is performed to the extracted peripheral blood mono-nuclear cell DNAs of a patient, the primers of the glutathione S-transferase M3 methylation (M) specificity and non-methylation (U) specificity are used to perform methylation-specific polymerase chain reaction (PCR). According to the amplification product, the methylation state can be judged and the clinician can judge the course of severe hepatitis and guide the treatment. |
