abstract | The invention discloses a preparation method of (R) or (S)-2-aminomethyl tetrahydropyrrole shown in D, which comprises the steps of: carrying out acylamino reduction reaction on compound B under the action of sodium borohydride and iodine in organic solvent, and obtaining compound C; and (2) removing tert-butoxycarbonyl protective group of amino of the compound C obtained in the step (1) under the action of trifluoroacetic acid in the organic solvent, and obtaining compound D, wherein * represents absolute configuration R or S of chiral carbon. The preparation method is fewer in reaction steps, simple in post process, lower in cost and higher in yield, and can avoid racemization of the product, thus obtaining the high-purity product with the optical activity purity more than over 99%, and being especially applicable to pilot amplification or industrialized production. |
