abstract The invention relates to a hydroxyproline structure-containing HIV-1 protease inhibitor as well as a preparation method and application thereof, belonging to the field of chemical drugs. A compound has the structure shown as the general formula 7 in the description. The preparation method comprises the following steps of: combining a Boc-protecting amine chiral alcohol compound as a raw material with a hydroxyproline methyl ester with hydroxyl being activated by a DSC (Dynamic Stability Control) reagent to generate carbamate, then hydrolyzing under alkali conditions to obtain corresponding carboxylic acid, carrying out condensation reaction with different amines to generate a series of amide derivatives, removing a Boc-protecting group and reducing nitro to synthesize a series of novel hydroxyproline structure-containing HIV-1 protease inhibitors. The target compound has strong HIV-1 protease inhibition activity and is used for preparing anti-AIDS drugs.
