abstract | The invention discloses an anti-uPAR humanized antibody, a coding gene and application thereof. The amino acid sequence of a heavy chain variable region of the antibody is shown as a sequence 3 in a sequence table, and the amino acid sequence of a light chain of the antibody is shown as a sequence 1 in the sequence table. Experiment results confirm that the antibody has good binding activity (affinity is 1.74*10-8 mol/L) and capability of inhibiting the growth and migration of tumor cells. The humanized antibody can be bound better to uPAR so as to guarantee anti-tumor effect thereof. A method for preparing the antibody can simultaneously express the light chain and a heavy chain and ensure that the expression ratio of the light chain to the heavy chain is closer to 1:1 so as to produce the antibody with double chains mutually matched in a higher ratio. In summary, the antibody and the preparation method thereof can have broad application prospects in the field of preventing and/or treating tumors. |
