abstract It is intended to provide a method for amplifying a target sequence by suppressing the inhibition of amplification reaction in nucleic acid amplification in the presence of a probe. In the amplification of a target sequence, as a probe to be allowed to coexist, a probe composed of a base sequence in which when a double-stranded nucleic acid of the probe and a strand complementary to the probe is formed, a melting temperature of the double-stranded nucleic acid is equal to or lower than a reaction temperature of the elongation reaction is used. If such a probe is present, for example, annealingof a primer or an elongation reaction is not easily inhibited due to the presence of the probe, and a target sequence can be sufficiently amplified. Accordingly, when polymorphism of locus to be detected in the target sequence is analyzed by Tm analysis or the like, high reliability can be achieved.
