abstract | A vector has a nucleic acid operably linking a promoter, an intron, a secretory leader sequence encoding nucleic acid, a human betacellulin (BTC) encoding nucleic acid, or functional fragment thereof, and a polyadenylation signal sequence, wherein expression of BTC produces a secreted, mature BTC. A method of treating or preventing diabetes includes administering an effective amount of a viral particle having this vector. Also provided is a vector having a nucleic acid operably linking a cytomegalovirus (CMV) promoter and enhancer region, a ss-globin chimeric intron, an albumin leader sequence encoding nucleic acid, a human betacellulin (BTC) encoding nucleic acid, or a functional fragment thereof, and an SV40 polyadenylation signal sequence, wherein expression of BTC produces a secreted, mature BTC. A host cell containing the vectors of the invention are also provided. |
