abstract | Anti-inflammatory substd-phenylakanoic acids and derivs. M3A. Arne new cpds. of formula (I): (where X = O or S; Y1 = H, OH, amino, NO2, halogen, Me, Et, 1-3C alkoxy, MeSO2, MeSO2NH, CF3, AcNH, MeS or PhO; R1 = H, 1-5C alkyl, 2-5C alkenyl or alkynyl or 3-6C cycloalkyl; and when PhX is para, n=0; Y2 = H, OH, halogen, 1-5C alkyl, 1-3C alkoxy (neither Y1 or Y2 is H when R1 is H or Me); Z = (a) COOR2 where R2 = H, 1-5C alkyl, di(1-3 C alkyl) amino-(1-4C) alkyl or an alkali or alk. earth metal, NH4 or substd. NH4; (b) CONR3R3 where R3 = same or diff. H, OH, 1-5C alkyl, cyclopropylmethyl or CH2COOR2; (c) CH2OR4 where R4 = H, 1-5C alkyl, Ac, EtCO, NH2CO2, NHMeCO2, Me2NCO2 or 3-6C alkoxy-alkyl; (d) tetrazol-2-y1; and when PhX is meta, n = 0-3; Y2 beta H, OH, halogen, Me or Et (but n = H when Y1 = OH or 1-3C alkoxy); R2 = 2-(3-phenoxyphenyl)propyl as well as above; Z = (e) when n = 1-3, NR5R5 where R5 = (1) H, 1-5C alkyl, cyclopropyl or cyclopropyl-methyl, or (2) Ac, EtCO, NHMeCO2 or NMe2CO2). (I) also have mild analgesic and antipyretic props. similar to aspirin; and some cpds. potentiate alpha-d-propoxyphene morphine and codeine. - Pref. cpds. include 2-(opt. substd.-3 and 4-phenoxyphenyl)-acetic and propionic acids and propylamines and prepn. is by known methods. |
