abstract | Pyrazolo (3, 4-b) pyridines of formula (I):- where R1 = H or alkyl, preferably CH3; R2 = H or alkyl; R3 = OH, halogen, NH2, or SH. Antimetabolites of considerable interest. Inhibitors of DNA and RNA synthesis. Intermediate (II) may or may not be isolated. A mixture of 5.18g. (III; R1= C6H5CH2), 4.28g. ethyl acetoacetate, and 15ml. acetic acid was refluxed for 4 hrs., cooled, and treated with 100 ml. water to precipitate 6.58g. (I; R1=C6H5CH2, R2=CH3, R3=OH), m. 228-9 deg. |
