abstract | (A) Cpds. of general formula (I) R' = (1-5C) alkyl, (2-5C) alkenyl, (is not > 7C) cycloalkyl, or (is not > 7C) cycloalkenyl R2 = Me or Et (B) Acid addn. salts of I. Oral and parenteral hypoglycaemics. Dose. 100-500 mg/d. (a) N'-(1-methyl-2-t.butylaminoethyl)-N4-acetyl-sulphanilamide (32.7 g.) was dissolved in 2N-NaOH (100 ml), BrCN (10.6 g.) added at 20-30 deg., stirred 30 mins., filtered, and recryst. from DMF, giving the acetanilide, m.p.243-4 deg. (b) This (35.2 g.) was added to 50% H2SO4 (125 g.) at 40 deg., stirred 2 h. at 50 deg., poured into H2O (750 ml), 10N-NaOH added to pH 6.3, stirred 15 mins. with charcoal (2 g.), filtered (Hyflo), basified (10N-NaOH), filtered, washed, dried, and recryst. from EtOAc, giving I, m.p.145-6 deg. |
