abstract | (A) Oxazoloisoquinolines (I) and their salts. - R, R1-3 = H, F, Cl, CF3; excluding o-(CF3)2 - R4 5 = H, F, Cl, 1-4C straight chain alkyl - R6 = H, 1-4C straight chain alkyl - Among R, R1-5 not more than 3 is not = H - (B) Intermediates (II), (VI) and (VII), below. - As antidepressants. Some are anorexigenic ("B isomers"). - (a) Condensation of 1-cyano-2-benzoyl-1, 2-dihydroisoquinoline and p-chlorobenzaldehyde in THF in the presence of PhLi at -10 deg. and saponification of the product with aq. alc. KOH yields alpha(p-chlorophenyl)-isoquinoline-1-methanol, m.p. 98-103 deg. - (b) Redn. with PtO2-H2 (Parr bomb) in AcOH gives the 1,2,3,4-tetrahydro-cpd., m.p. 97-101 deg. (pentane) - (c) This (7.3 g.) and NaOAc (4.4 g.) in MeOH at 5 deg. is treated with a soln. of CNBr (2.8 g.) and the mixt. stirred at room temp. for 12 hrs. Working up yields 1-(p-chlorophenyl)-3-imino-1, 5, 6, 10b-tetrahydro-3H-oxazolo(4, 3-a)isoquinoline (isomer A), m.p. 137-8 deg. (pentane-benzene), HCl salt, m.p. 160-8 deg., maleate, m.p. 168-9 deg. |
