abstract The present invention provides methods and reagents for use in the amplification of nucleic acids. Amplification and detection carried out using probe oligonucleotides containing an N2-benzyl-dG nucleotide can result in less non-specific amplification involving the probe compared to amplification carried out using unmodified oligonucleotides, presumably because the presence of a bulky N2-benzyl-dG minor groove binder in a template prevents extension of the complementary strand by a DNA polymerase.
