abstract | The present invention generally pertains to methods for detecting the presence or absence of a particular nucleic acid sequence. The invention relates to incorporating a detector into a target nucleic acid, adding an oligonucleotide probe, polymerase enzyme and inhibitor to the reaction, and detecting interference of the oligonucleotide probe with the inhibitor as an indication of the presence of a particular target nucleic acid sequence. A detector oligonucleotide conjugated to a fluorophore or quencher is incorporated at the 3' end of a target nucleic acid, and is annealed to an inhibitor oligonucleotide conjugated to the corresponding quencher/fluorophore such that fluorescence of the fluorophore is quenched. An oligonucleotide probe anneals to the target nucleic acid, and serves as a primer for extension by a DNA polymerase to generate a strand complementary to the target nucleic acid, which displaces the inhibitor oligonucleotide to generate fluorescence indicating the detection of the target nucleic acid. |
