abstract | Disclosed herein are detectably-labeled probes for detecting a target nucleic acid in a sample. The probes comprise an anchor domain and a reporter domain linked by a non-nucleoside linker. The anchor and reporter domains are less than 50% complementary and neither forms a stem loop in the absence of target. The probe is not extendible by polymerase and the reporter domain comprises a non-natural nucleotide to increase binding specificity. The linker is linked to the anchor domain within 2 nucleotides of its 3' end and to the reporter domain within 2 nucleotides of its 5' end. The anchor domain is not linked to a label. Also disclosed are methods, reaction mixtures and kits employing the probes disclosed herein. |
