abstract | The present invention relates to compositions and methods for analyzing and modulating (e.g., enhancing or inhibiting) protein-protein interactions. In particular, compositions and methods of the present invention find use in identifying, reconstituting and analyzing protein-protein interactions, identifying binding subunits, and drug screening. The system of the invention utilizes first and second fusion proteins, the first fusion protein comprising a Twin Arginine translocation signal sequence and a first test polypeptide, and the second fusion protein comprising a marker protein and said second test polypeptide. Cells expressing the fusion proteins are subjected to selective pressure to identify interactions between the first and second test polypeptides. |
