abstract This invention provides methods and reagents for determining or predicting responses to cancer therapy, as well as dual therapy treatments. The methods involve identifying mammalian tumors that can be treated with dual mTOR pathway inhibitor and EGF pathway inhibitor therapy, comprising the step of assaying a sample from the tumor to detect expression and/or phosphorylation of polypeptides of the EGF pathway and of the mTOR pathway. EGF pathway polypeptides may include ERK and MEK, while mTOR pathway polypeptides may include mTOR and/or HIF-I alpha .
