abstract | The present invention relates to a series of substituted bicyclic[3.1.0] heteroaryl amides of the Formula I, (see formula I) wherein: R1 is an optionally substituted heteroaryl; R2, R3 and A independently represent H or optionally substituted alkyl; Q represents -(CH2)n- or -(CH2)m-O- where n is 1, 2, 3 or 4 and m is 2, 3 or 4; Z represents aryl, alkyl or cycloalkyl; R4 and R5 each independently represent H, halogen or optionally substituted alkyl, aryl, aryloxy, alkoxy, heterocycloalkyl, or cycloalkoxy; Y represents -R6, -(CH2)o-R6, -C(R6)3 or -CH(R6)2 where o is 1, 2 or 3; R6 represents H, aryl, alkyl, cycloalkyl, bicycloalkyl, tricycloalkyl, heterocycloalkyl, heteroaryl, C(=O)NR7R8 or -C(=O)OR7 wherein said R6 groups can be optionally substituted with one or more X groups; X represents -OH, alkoxy, -NR11R12, -SO2R10, -C(=O)R10, halogen, cyano, alkyl, alkoxyalkyl, heteroaryl, aryl, aryloxy, benzyl or hydroxyalkyl; R7 and R8 independently represent H, alkyl, cycloalkyl, heterocycloalkyl, hydroxyalkyl or alkoxyalkyl wherein R7 and R8 may be substituted by one or more X groups or R7 and R8 together with the nitrogen in which they may be attached may form a heterocycloalkyl group optionally substituted by one or more X groups; R10 represents alkyl, cycloalkyl, heterocycloalkyl, hydroxyalkyl, heteroaryl or alkoxyalkyl and R11 and R12 independently represent H, alkyl, cycloalkyl, heterocycloalkyl, hydroxyalkyl, heteroaryl, or alkoxyalkyl, that exhibit activity as glycine transport inhibitors, their pharmaceutically acceptable salts, pharmaceutical compositions containing them, and their use for the enhancement of cognition and the treatment of the positive and negative symptoms of schizophrenia and other psychoses in mammals, including humans. |
