abstract The present invention concerns chimeric molecules that contain a preferential polypeptidic region, consisting of a specific affinity for the binding to specific DNA sequences, of a preferential polypeptidic region consisting of a DNA modifying activity, and this chimeric molecule is capable to cross biological membranes due to the presence of a region that contains delivery activity. The invention contains further the isolated polynucleotides that code for the chimeric molecules of the invention if they are as such entirely or partially of polypeptide nature. In another embodiment, based on the activities of the polypeptides contained in the invention to interfere with key points of the cell-cycle regulation and the cellular checkpoints due to their introduction of DNA double strand breaks, the invention contains various procedures that are characterized by the use of said polypeptides of the invention for cells in vivo and provides an activity for the modification of specific sites of the DNA contained in a cell. The invention also contains procedures that use the chimeric molecules of the invention to screen for new delivery activities or combinations of delivery activities. The invention further provides for the therapeutic use of said compositions as anti-prolierative, anti-neoplastic, antibiotic, antiparasitic or antiviral agents.
