abstract | The invention relates to a method for exhaustive detection of bacteria genes pathogens, in particular deNm, expressing a desired phenotype, characterized in that: - a mutant bank generated from a strain is used bacterial given so that at least 70% of the nonessential genes, and in particular at least 80%, or even more than 90%, are mutagenized by insertion of a transposon in a reading phase, - then the mutants, either individually or in pools, with an environment, such as medium, animal or cells, capable of interacting with mutant bacteria expressing the desired phenotype, - we recover, in the case of use of pools, the bacteria not having interacted with the desired phenotype, -we identify the mutated genes of these bacteria and we verify their implication in said phenotype. Applications, in particular, as targets antipathogenicity, consisting in inhibiting the growth of Neisseria meningitidis in vivo in serum, to develop antibiotics, to screen and manufacture drugs to open the barrier hematoencephalic to active ingredients, and develop vaccines. |
