abstract Provided are substrates and assays for the identification of caspase activators and inhibitors. The substrates are fusion proteins comprising two green fluorescent proteins (GFPs) with a linker peptide comprising at least one caspase cleavage site. The intact fusion protein exhibits fluorescent resonance energy transfer (FRET) between the GFPs. Following caspase cleavage of the linker peptide, the two GFPs become separated and FRET is diminished.
