abstract | To search human Cds1 (hCds1) being homologous with yeast cell cycle regulatory factors scCds1 and spCds1, GenBank EST data base is retrieved based on fission yeast spCds1 sequence and baker's yeast scCds1 sequence, thereby finding a homologous molecule (dCds1) of Drosophila which is significantly homologous with spCds1 and sdCds1. Then the full-length gene thereof is isolated. It is confirmed that this gene is expressed strongly in testis and, moreover, widely expressed in other tissues. A protein encoded by the thus isolated gene is prepared as a recombinant protein and its kinase activity is detected in vitro. As a result, it is found out that this protein efficiently phosphorylates cdc25 and histone H1. |
