abstract | A gene encoding a hybrid protein consisting of ALA-S yeast and a N-terminal transit sequence for the small carboxydismutase sub-unit is introduced into the genim of tobacco (nicotania tabacum) and a chlorophyll-poor tobacco transformant which expresses glutamate-1-semialdehyde-transminase (GSA-AT) antisense RNA. The expression of ALA-S in the GSA-AT transgenic antisense strain has produced green co-transformants, the chlorophyll content of which is similar to that of the wild type, while the wild-type transformants have shown no phenotypical changes. The capacity of plants to synthetize ALA and chlorophyll is increased, thereby demonstrating the contribution of ALA-S to the ALA supply required for the chlorophyll synthesis. The transgenic plants produce chlorophyll in the presence of gabaculine, a GSA-AT inhibitor. The permanent RNA and protein content of GSA-AT have been reduced, as well has the enzymatic activity of plants expressing ALA-S. The additional synthesis of ALA also explains the expression of GSA-AT in the C5 way. |
