abstract | New farnesyl transferase inhibitors of general formula (I), their preparation and pharmaceutical compositions containing them. In the general form (I), R1 represents YS-A1- (Y = hydrogen atom, remainder amino acid, fatty acid residue, alkyl or alkoxycarbonyl radical or a radical R4-S- in which R4 represents an alkyl radical containing 1 to 4 carbon atoms optionally substituted by a phenyl radical or a radical of general formula (II) in which A1, X, X1, Y1, R'1, R2, R'2 and R are defined as below, and A1 = alkylene radical containing 1 to 4 atoms of carbon optionally substituted in .alpha. of the grouping> C (X1) (Y1) by a amino or alkylamino, dialkylamino, alkanoylamino or alkoxycarbonylamino), X1 and Y1 each represents a hydrogen atom or together form with the carbon atom to which they are linked a group > C = O, R'1 represents hydrogen or an alkyl radical containing 1 to 6 atoms of carbon, X represents an oxygen or sulfur atom, R2 represents a alkyl, alkenyl or alkynyl radical containing 1 to 6 carbon atoms optionally substituted by hydroxy, alkoxy, mercapto, alkylthio, alkylsulfinyl or alkylsulfonyl, it being understood that when R2 represents a alkyl radical substituted by a hydroxy radical, R2 can form with the carboxy radical in .alpha. a lactone, R'2 represents hydrogen or a radical alkyl containing 1 to 6 carbon atoms, and R represents a hydrogen atom or an optionally substituted alkyl radical or a phenyl radical optionally substituted, it being understood that the radical (a) is in position -3 or -4 from the naphthyl nucleus. These new products have properties anticancer. |
