abstract | An expression system for producing an enzyme, and a method for preparing same, are disclosed. The system includes a promoter nucleotide sequence, a nucleotide sequence coding for a secretion signal, a nucleotide sequence coding for a mature enzyme and a terminator nucleotide sequence. The expression system is particularly useful for producing a Pseudomonas lipase. Said expression system may be inserted into a vector. A Pseudomonas wisconsinensis strain transformed by the expression vector is used as the host strain. The expression system may also include a nucleotide sequence coding the Pseudomonas wisconsinensis lipase modulator and/or a nucleotide sequence coding for the GPW protein included in the Pseudomonas wisconsinensis lipase operon. The Pseudomonas wisconsinensis lipase modulator that is involved in effective lipase expression and aids the formation of an active lipase is also disclosed. Furthermore, the GPW protein that appears also to be involved in effective lipase expression is disclosed. The resulting lipase may be used in detergent compositions. |
