abstract | Provided herein is LTC4 synthase, in substantially free form, having a molecular weight of 18 kDa as determined on silver stained SDS-polyacrylamide denaturing gel, said LTC4 synthase having the partial amino-terminal sequence: M K D E V A L L A A V T L L G Met-Lys-Asp-Glu-Val-Ala-Leu-Leu-Ala-Ala-Val-Thr-Leu-Leu-Gly- 15 V L L Q A G F S L Q V I S A R Val-Leu-Leu-Gln-Ala-Gly-Phe-Ser-Leu-Gln-Val-Ile-Ser-Ala-Arg- 30 X A F R V [Xaa]-Ala-Phe-Arg-Val-wherein X is I (Ile) or R (Arg). The enzyme is useful in the identification of specific LTC4 synthase inhibitors, in the biosynthetic production of LTC4, and as an antigen for the production of polyclonal and monoclonal antibodies, which in turn could be used to neutralize the enzyme and thereby prevent the production of the pro-inflammatory TC4. |
