abstract | RNA amplification process, comprising steps of denaturing RNA by heating a solution containing it, synthesize a first strand of cDNA to from an appropriate primer in the presence of a reverse transcriptase, denature the heteroduplex formed, synthesize the second strand of cDNA from a second primer in the presence of DNA polymerase, then to be submitted cDNA obtained in number amplification cycles sufficient. Thanks to the discovery that some reversible transcriptases not known to be thermostable are capable, when used in quantity sufficient, to resist denaturing treatment of RNA by heat at a temperature greater than or equal at 60 ° C, and even to be able to operate, after this treatment, at high temperatures corresponding to sufficiently discriminating conditions, this process is characterized by the fact that said RNA solution contains from the start, all necessary reagents and solvents at said stages and that all of the stages are carried out in the same container without adding reagents or solvents. It is thus possible to operate, in satisfactory conditions, in a single step of handling, which in particular avoids risks of contamination. Application in particular to the diagnosis of infections bacterial or viral, and genetic disorders, as well as cell typing. |
